How do I connect an audio sound interface port info Spartan3E dev kit?
Or is there any other board that has readily available port?

There are many different S3E dev kits.
Audio port could also mean very different things:
-mic level input
-line level analog input or output (+4dBu or -10dBV)
-digital inputs or outputs, e.g. AES/EBU, S/PDIF, ADAT
The Xilinx ML40x and ML50x boards have an audio codec and associated connectors. More information is available in the respective user guides.
If you are using the Xilinx/Digilent Spartan-3E Starter Kit, I have seen a few approaches:

-use an analog PMOD (peripheral module) board, e.g.

I haven't personally tried these approaches yet.
This question is also likely better suited for the "Boards and Kits" board since it is not Spartan silicon specific. Just an FYI for next time.

Thanks for your recommendations. Yes I'm going to connect a microphone.
Now that I've got the Xilinx spartan3E evaluation kit, and the PMOD mic recommended. the PMOD mic has an on-board preamp, 12-bit ADC and outputs the signal into the Xilinx spartan3E eval kit's analogue capture circuit which will go through the on-board 14-bit ADC.
Will there be any conflict (i.e. the analog capture circuit recognizes the incoming Vin A/ VinB signal as analogue signal and re-do the A/D conversion? If yes, is there anyway to bypass the 1st or second ADC.
